Bilan de la toxicité aiguë
Le S-métolachlore est faiblement toxique quelle que soit la voie d'exposition. Il est légèrement irritant pour les yeux mais faiblement pour la peau. Il est un sensibilisant cutané.

Le s-métolachlore n'appartient pas à une classe de produit connu pour causer des effets endocriniens. Il n'y a eu aucune évidence d'effet sur la fonction endocrinienne dans les études sur la reproduction ou le développement. De plus, les examens histologiques des organes endocriniens chez le chien, le rat ou la souris n'indiquent pas que le système endocrinien est affecté par le s-métolachlore même à la dose maximale tolérée la vie durant.

Les études sur le développement prénal chez les rats et les lapins n'ont pas révélé d'évidence de sensibilité qualitative ou quantitative accrue des fœtus comparativement aux mères. Aucun effet significatif n'a été observé dans la plupart des études même à la plus forte dose testée. Les résultats indiquent que le s-métolachlore est ni embryotoxique ou tératogène chez l'une ou l'autre des espèces même à des doses toxiques maternelles.

Bilan des effets à long terme
Le s-métolachlore n'a pas causé de toxicité particulière dans les études chroniques chez les animaux de laboratoire l'effet le plus marqué étant une diminution de gain de poids corporel. Toutefois, il est classé comme un cancérogène possible chez l'humain pour avoir causé une augmentation de nodules néoplasiques et de carcinomes hépatocellulaires à une dose élevée chez les femelles des rats. Aucun effet sur le développement, la reproduction ou sur le système endocrinien n'a été signalé. Le s-métolachlore n'est ni génotoxique ni neurotoxique.
